Bioluminescence Resonance Energy Transfer
The technique of BRET ( Bioluminescence Resonance Energy Transfer ) appeared these last years like a novel method making it possible to study the interactions Protéine - protein in a dynamic way. In the technique of BRET, one of partners is amalgamated with a Luciférase (Renilla luciférase), tandis that the other partner is amalgamated with a fluorescent protein (e.g. YFP, Fluorescent Yellow Protein). The luciférase is excited by addition of its substrate, the Coelentérazine. If the two partners interact, a transfer of energy is done by resonance, of the luciférase towards the YFP, and a fluorescent signal, emitted by the YFP, can be detected.
